On Tuesday 4 June 2024, A. van Hemelryk will defend the doctoral thesis titled: ‘Of Men, Mice and Organoids: a Story Tailored for Prostate Cancer‘.
Brief summary of the doctoral thesis:
Castration-resistant prostate cancer (CRPC) remains a lethal disease due to the inevitable occurrence of therapy resistance. There is a pressing need for preclinical models that accurately capture the ever-evolving clinical landscape of treatment-resistant CRPC. These models will help understand the mechanisms of resistance and will allow to explore new drugs or drug combinations in late-stage disease. The relevance of patient-derived xenograft (PDX) models - which closely mimic human tumors - is well-established, but more easy-to-manipulate models are crucial to allow early bench-to-bedside translation of new drug discoveries and guidance of therapeutic decision making. This thesis aimed to advance preclinical CRPC research and move forward its clinical translation in two interlinked ways: first, by providing the research field with new, relevant models of heavily pretreated CRPC (part I), and secondly, by implementing and optimizing in vitro technologies for assessing drug response in three-dimensional (3D) organoid models (part II).
