Brief summary:
Hypertension is a major global health concern, significantly contributing to cardiovascular morbidity and mortality. Central to its regulation is the renin-angiotensin system (RAS), with angiotensinogen (AGT) as a pivotal precursor for angiotensin II, a potent vasoconstrictor. This thesis explores the therapeutic potential of liver-targeted small interfering RNA (siRNA) therapies designed to suppress AGT production, offering a novel approach to modulate the RAS and reduce blood pressure. In addition to evaluating the efficacy of AGT suppression, the work investigates the reversal mechanisms involved and compares various anti-hypertensive drug classes and combinations, aiming to identify the most effective strategies for long-term blood pressure control.
