With the introduction of immune checkpoint inhibitors (ICIs) and the first Kirsten rat sarcoma virus (KRAS) G12C inhibitors, significant strides have been made in the management of patients with advanced KRAS-mutated non-small cell lung cancer (NSCLC). However, outcomes in this patient population remain modest compared to outcomes in patients with other oncogenic drivers such as epidermal growth factor receptor (EGFR) or anaplastic lymphoma kinase (ALK) alterations. The considerable heterogeneity of KRAS-mutated NSCLC, likely due to its strong association with smoking, presumably contributes to this discrepancy. In this thesis, we aimed to provide insights into integrating novel technologies, such as circulating tumour DNA (ctDNA), and advancements in drug development into daily clinical practice to improve outcomes for patients with KRAS-mutated NSCLC.
